Fostriecin was isolated for the first time in 1983 from the fermentation broth of an actinomycetes collected in soil samples from Brazil. It belongs to natural lactone compounds, and their producing bacteria are Streptomycespulveraceus. This product has good anti-tumor activity, compared with 17 kinds of clinically applied drugs, it is one of the compounds with the best activity. In vitro experiments have shown that fostrixine can inhibit topoisomerase Ⅱ and protein phosphatase 2A (ProteinPhosphatase2A, PP2A), but its in vitro cytotoxicity mainly inhibits topoisomerase Ⅱ through a non-DNA cleavage mechanism. It shows obvious anti-tumor activity on mouse leukemia L1210 and P388 cells cultured in vitro and human lung cancer, breast cancer and ovarian cancer cells. At present, the clinical phase I study of Fostriecin has been completed. During the phase I clinical trial, it was found that it can prevent tumor recurrence, but the plasma half-life is short; the main adverse reactions are vomiting, fever and mild fatigue. The clinical trial was interrupted due to the detection of an 8% impurity in the natural product. At present, the chemical synthesis of fostrixin has been completed, and clinical trials are expected to continue.
